清雍正二年（1724年）凤阳府泗州升为直隶，领盱眙、天长、五河三县①。乾隆四十二年（1777）凤阳府省虹县入泗州，泗州辖境扩大。此后至清末泗州之所未有更变。
①《清嘉庆一统志》第42册，卷125《凤阳府一建置沿革》：“凤阳府，……（洪武）七年改曰凤阳府，始移今治。直隶南京。本朝初属江南布政使司。康熙六年分属安徽省。旧领州五、县十三。雍正二年升颍州直隶，以颍上、霍丘二县属之。升亳州直隶，以太和、蒙城二县属之。升泗州直隶，以盱眙、天长、五河三县属之。十一年分寿州地，增置凤台县。乾隆十九年裁临淮县，并归凤阳县。四十二年裁虹县，以地属泗州直隶州。领州二、县五。”",
